KRAS G12D

Description

RMC-9805 is a RAS(ON) inhibitor specifically designed to target KRAS G12D-mutated solid tumors, including colorectal cancer. This compound represents a novel approach to directly inhibiting mutant KRAS proteins in their active GTP-bound state. RMC-9805 is being developed for patients with KRAS G12D mutations, which occur in approximately 12-15% of colorectal cancers and have historically been considered "undruggable" targets.

Mechanism of Action

RMC-9805 functions as a RAS(ON) inhibitor that directly binds to and inhibits the KRAS G12D mutant protein in its active, GTP-bound conformation. This mechanism differs from traditional KRAS G12C inhibitors by targeting the protein while it is in the "on" state rather than the inactive GDP-bound state. By blocking active KRAS G12D signaling, the drug disrupts downstream oncogenic pathways including RAF-MEK-ERK and PI3K-AKT cascades that drive tumor growth and survival.
