Description

Relatlimab is a monoclonal antibody immunotherapy agent that targets the LAG-3 (Lymphocyte Activation Gene-3) immune checkpoint receptor. In colorectal cancer, it is being evaluated specifically for microsatellite stable (MSS) tumors, which typically do not respond well to standard PD-1/PD-L1 checkpoint inhibitors. Relatlimab is administered in combination with nivolumab (anti-PD-1) to enhance immune system activation against cancer cells in this challenging patient population.

Mechanism of Action

Relatlimab blocks the LAG-3 receptor on T cells, which normally serves as an immune checkpoint that suppresses T cell activation and proliferation. By inhibiting LAG-3, relatlimab removes this brake on the immune system, allowing T cells to maintain their anti-tumor activity for longer periods and potentially overcome immune suppression in the tumor microenvironment.
