Description

Fianlimab is a monoclonal antibody immunotherapy agent that targets the LAG-3 (Lymphocyte Activation Gene-3) immune checkpoint receptor. In colorectal cancer treatment, fianlimab is being evaluated in combination with cemiplimab (anti-PD-1 antibody) for patients with both microsatellite instability-high (MSI-H) and microsatellite stable (MSS) tumors. The combination approach aims to enhance anti-tumor immune responses in patients with localized, locally advanced, and resectable non-metastatic colorectal cancer.

Mechanism of Action

Fianlimab blocks the LAG-3 receptor, which is an inhibitory checkpoint protein expressed on activated T cells and other immune cells. By inhibiting LAG-3, fianlimab prevents the suppression of T cell activation and proliferation, thereby enhancing the body's immune response against cancer cells. When combined with PD-1 inhibitors like cemiplimab, this dual checkpoint blockade can provide synergistic anti-tumor activity by targeting multiple immune suppressive pathways.
