Description

Etrolizumab is a humanized monoclonal antibody that selectively targets the ?7 integrin subunit, blocking the ?4 7 and ?E 7 integrins involved in inflammatory cell trafficking to the gastrointestinal tract. While primarily developed for inflammatory bowel diseases like ulcerative colitis, this agent is not established as a standard treatment for colorectal cancer. The drug's mechanism of blocking gut-specific lymphocyte homing may have theoretical applications in CRC-related inflammation, though clinical evidence for oncologic efficacy remains limited.

Mechanism of Action

Etrolizumab binds specifically to the ?7 integrin subunit, preventing the interaction between ?4 7 integrin on circulating lymphocytes and MAdCAM-1 on gut endothelial cells. This selective blockade reduces inflammatory cell migration into intestinal tissues while preserving systemic immune function, as it does not affect ?4 1 integrin-mediated trafficking to other organs.
