Description
Aflibercept is a recombinant fusion protein that functions as an anti-angiogenic agent used in combination with FOLFIRI (irinotecan, 5-fluorouracil, and leucovorin) for the treatment of metastatic colorectal cancer. It is specifically indicated for patients whose disease has progressed following treatment with an oxaliplatin-containing regimen. Aflibercept works by blocking the formation of new blood vessels that tumors need to grow and spread.
Mechanism of Action
Aflibercept acts as a soluble decoy receptor that binds to and neutralizes vascular endothelial growth factor-A (VEGF-A), VEGF-B, and placental growth factor (PlGF). By sequestering these pro-angiogenic factors, it prevents them from binding to their native receptors on endothelial cells, thereby inhibiting angiogenesis and reducing tumor vascularization and growth.
