Zibotentan (** Generic name only (development discontinued) **)
Other Medications
Description
Zibotentan is an experimental drug that was studied in combination with standard chemotherapy (FOLFIRI: irinotecan, fluorouracil, leucovorin) for metastatic colorectal cancer that had progressed after treatment with oxaliplatin-based chemotherapy. The goal of the clinical trial was to see if adding zibotentan to FOLFIRI would improve progression-free survival (the time before the cancer starts growing again) compared to FOLFIRI alone. The trial also aimed to assess the safety and feasibility of this treatment combination. Zibotentan was given orally for 14 days, followed by a period of zibotentan alone for patients whose cancer stabilized. The trial also collected samples to study potential biomarkers related to the drug's effect. Development of zibotentan has been discontinued.
Mechanism of Action
Zibotentan is an antagonist of the endothelin A receptor (ETAR). Endothelins are substances in the body that can cause blood vessels to narrow. By blocking the ETAR, zibotentan may affect blood vessel function, potentially impacting tumor growth and spread. The specific role of ETAR in colorectal cancer and the exact mechanism by which zibotentan might affect it are areas of ongoing research.
