Description

Venetoclax is an oral targeted therapy belonging to the BCL-2 inhibitor class that selectively targets the BCL-2 protein to induce apoptosis in cancer cells. While primarily approved for hematologic malignancies, venetoclax is being explored in colorectal cancer through precision medicine approaches and combination trials. The drug's use in CRC is based on the rationale that some colorectal tumors may have dependencies on BCL-2 family proteins for survival and resistance to conventional therapies.

Mechanism of Action

Venetoclax functions as a selective, orally bioavailable small-molecule inhibitor of the BCL-2 anti-apoptotic protein. By binding to and inhibiting BCL-2, the drug disrupts the protein's ability to prevent programmed cell death, thereby restoring the natural apoptotic process in cancer cells. This mechanism is particularly relevant in tumors that overexpress BCL-2 or have defective apoptotic pathways.
