Description
Vandetanib is an oral multi-targeted tyrosine kinase inhibitor that has been evaluated in combination with standard chemotherapy regimens for colorectal cancer treatment. This small molecule inhibitor blocks multiple growth factor receptors simultaneously, including EGFR, VEGFR, and RET pathways. In colorectal cancer, vandetanib has been combined with FOLFOX and FOLFIRI chemotherapy backbones, as well as with capecitabine, oxaliplatin, and bevacizumab regimens for metastatic disease.
Mechanism of Action
Vandetanib works by inhibiting multiple receptor tyrosine kinases that drive tumor growth and angiogenesis. It primarily targets the epidermal growth factor receptor (EGFR), vascular endothelial growth factor receptors (VEGFR-2 and VEGFR-3), and RET proto-oncogene receptor, thereby blocking key signaling pathways involved in tumor cell proliferation, survival, and blood vessel formation.
