Description

UFOX is a modified chemotherapy regimen used in the treatment of colorectal cancer that represents a variation of the standard FOLFOX protocol. This regimen combines fluorouracil, leucovorin, and oxaliplatin in an altered dosing schedule or formulation compared to traditional FOLFOX-4. UFOX is employed as a backbone chemotherapy regimen that can be combined with targeted agents like cetuximab for enhanced therapeutic efficacy in metastatic colorectal cancer patients.

Mechanism of Action

The regimen works through complementary cytotoxic mechanisms targeting DNA synthesis and repair pathways in cancer cells. Fluorouracil acts as a pyrimidine antimetabolite that inhibits thymidylate synthase, while leucovorin enhances fluorouracil's efficacy by stabilizing the drug-enzyme complex, and oxaliplatin forms DNA crosslinks that prevent replication and transcription.
