Description
Tremelimumab is a fully human monoclonal antibody that targets CTLA-4 (cytotoxic T-lymphocyte-associated protein 4) for the treatment of various solid tumors including colorectal cancer. This immunotherapy agent works by blocking inhibitory signals that prevent T-cell activation, thereby enhancing the body's immune response against cancer cells. In colorectal cancer treatment, tremelimumab is primarily used in combination with other agents such as durvalumab or cabozantinib, particularly in patients with microsatellite instability-high (MSI-H) tumors or specific molecular profiles including KRAS mutations.
Mechanism of Action
Tremelimumab binds to CTLA-4 receptors on T-cells, blocking the interaction between CTLA-4 and its ligands CD80 and CD86 on antigen-presenting cells. This blockade prevents the inhibitory signals that normally downregulate T-cell responses, leading to enhanced T-cell proliferation, activation, and anti-tumor immune activity.
