Description

TNO155 is a small molecule inhibitor of SHP2 (Src homology 2 domain-containing protein tyrosine phosphatase 2) being evaluated in combination therapies for colorectal cancer. This compound targets tumors harboring BRAF V600E mutations and KRAS mutations, which are common driver mutations in colorectal cancer that lead to resistance to standard therapies. TNO155 has been studied in combination with KRAS G12C inhibitors like adagrasib, as well as with immunotherapy agents and CDK4/6 inhibitors in patients with advanced or metastatic disease.

Mechanism of Action

TNO155 inhibits SHP2, a protein tyrosine phosphatase that plays a critical role in multiple oncogenic signaling pathways including RAS/MAPK, PI3K/AKT, and JAK/STAT pathways. By blocking SHP2 function, TNO155 disrupts these growth-promoting signals that are often hyperactivated in BRAF and KRAS mutant tumors. This mechanism makes SHP2 inhibition an attractive approach for overcoming resistance to targeted therapies in RAS pathway-driven cancers.
