Description

Simvastatin is an HMG-CoA reductase inhibitor (statin) traditionally used to lower cholesterol that is being repurposed for colorectal cancer treatment based on its potential anti-tumor properties. In CRC, simvastatin is combined with standard chemotherapy regimens like FOLFIRI or XELOX plus bevacizumab to potentially enhance treatment efficacy in metastatic disease. The drug's cholesterol-lowering mechanism may disrupt cancer cell membrane function and cellular signaling pathways that promote tumor growth and metastasis.

Mechanism of Action

Simvastatin inhibits HMG-CoA reductase, the rate-limiting enzyme in cholesterol biosynthesis, which reduces cellular cholesterol production and affects membrane composition. In cancer cells, this mechanism may disrupt lipid raft formation and interfere with oncogenic signaling pathways, potentially leading to reduced cell proliferation, increased apoptosis, and decreased metastatic potential.
