Description

Ramucirumab is a fully human monoclonal antibody that blocks blood vessel formation in tumors. In colorectal cancer, it is approved for use in combination with FOLFIRI chemotherapy as second-line treatment for metastatic disease that has progressed after first-line therapy containing bevacizumab, oxaliplatin, and a fluoropyrimidine. It is also used in combination with TAS-102 (trifluridine/tipiracil) for patients with refractory metastatic colorectal cancer.

Mechanism of Action

Ramucirumab specifically binds to the vascular endothelial growth factor receptor 2 (VEGFR-2) on endothelial cells, preventing VEGF ligands from activating this receptor. By blocking VEGFR-2 signaling, it inhibits angiogenesis and reduces tumor blood supply, ultimately limiting tumor growth and metastasis.
