Description

Raltitrexed-PIPAC combines the antimetabolite chemotherapy drug raltitrexed with pressurized intraperitoneal aerosol chemotherapy (PIPAC) delivery for treating peritoneal metastases from colorectal cancer. This approach delivers raltitrexed directly into the peritoneal cavity as a pressurized aerosol, allowing for enhanced drug penetration into peritoneal tumor deposits while minimizing systemic exposure.

Mechanism of Action

Raltitrexed inhibits thymidylate synthase, a key enzyme required for DNA synthesis and cell division, leading to depletion of thymidine triphosphate and subsequent cancer cell death. The PIPAC delivery system uses pressurized aerosol application to improve drug distribution and penetration into peritoneal tissues compared to conventional intraperitoneal administration.
