Description
Panitumumab is a fully human monoclonal antibody that inhibits the epidermal growth factor receptor (EGFR) for treatment of metastatic colorectal cancer. It is specifically indicated for patients with RAS wild-type metastatic colorectal cancer, as tumors with KRAS or NRAS mutations do not respond to EGFR inhibition. The drug is used in combination with chemotherapy regimens like FOLFOX or FOLFIRI, or as monotherapy in patients who have progressed on fluoropyrimidine, oxaliplatin, and irinotecan-based chemotherapy.
Mechanism of Action
Panitumumab binds specifically to the extracellular domain of the human epidermal growth factor receptor (EGFR), blocking the binding of natural ligands like EGF and TGF-alpha. This prevents EGFR autophosphorylation and downstream signaling through pathways including RAS-RAF-MEK-ERK and PI3K-AKT, which normally promote cell proliferation, survival, and metastasis. By blocking these growth signals, panitumumab inhibits tumor cell growth and induces apoptosis in EGFR-expressing cancer cells.
