Description
Onvansertib is a small molecule polo-like kinase 1 (PLK1) inhibitor being developed for treatment of metastatic colorectal cancer, particularly in patients with KRAS mutations. The drug is administered in combination with standard chemotherapy regimens including FOLFIRI and bevacizumab or FOLFOX and bevacizumab for second-line treatment. Clinical trials have focused on its potential efficacy in RAS-mutated colorectal cancers, where limited targeted therapy options exist.
Mechanism of Action
Onvansertib selectively inhibits polo-like kinase 1 (PLK1), a serine/threonine kinase essential for cell cycle progression, particularly during mitosis. By blocking PLK1 activity, the drug disrupts spindle formation and chromosome segregation, leading to mitotic arrest and apoptosis in rapidly dividing cancer cells.
