Omeprazole (Prilosec, Losec, Zegerid)
Other Medications · Approved since 1989
Description
Omeprazole is a proton pump inhibitor (PPI) that reduces gastric acid production and is used as supportive care in colorectal cancer treatment. In the CRC setting, it serves primarily to prevent gastrointestinal toxicity from chemotherapy regimens, particularly oxaliplatin-induced complications, and to reduce gastric irritation when patients receive concurrent NSAIDs or aspirin for cancer prevention. Clinical applications include gastroprotection during aggressive chemotherapy protocols and as part of combination prevention strategies with low-dose aspirin.
Mechanism of Action
Omeprazole irreversibly binds to and inhibits the hydrogen-potassium ATPase enzyme (proton pump) located in gastric parietal cells. This action blocks the final step of gastric acid secretion, significantly reducing stomach acid production and creating a less acidic environment that protects the gastrointestinal tract from chemotherapy-related damage.
