Description

Mitomycin is an antitumor antibiotic chemotherapy agent derived from Streptomyces caespitosus that functions as an alkylating agent. In colorectal cancer treatment, it is primarily used in specialized applications including pressurized intraperitoneal aerosol chemotherapy (PIPAC) for peritoneal carcinomatosis and hepatic arterial infusion for liver metastases. Mitomycin may also be used in combination regimens for advanced or metastatic colorectal cancer, particularly in salvage therapy situations.

Mechanism of Action

Mitomycin acts as a bioreductive alkylating agent that becomes activated in hypoxic conditions typical of tumor environments. Once activated by cellular reductases, it forms DNA cross-links between complementary DNA strands, preventing DNA replication and transcription, ultimately leading to cell death.
