Description

Maraviroc is a CCR5 antagonist that blocks the C-C chemokine receptor type 5, originally developed and approved for HIV treatment. In colorectal cancer, maraviroc is being combined with PD-1 inhibitors specifically for microsatellite stable (MSS) metastatic colorectal cancer. This combination approach targets the CCR5 pathway to potentially enhance immune system activation in tumors that typically do not respond well to immunotherapy alone.

Mechanism of Action

Maraviroc selectively binds to and blocks the CCR5 co-receptor on immune cells, preventing CCL5 (RANTES) and other chemokines from binding. By inhibiting CCR5 signaling, maraviroc may reduce immunosuppressive signals in the tumor microenvironment and enhance T-cell infiltration, potentially making microsatellite stable colorectal cancers more responsive to checkpoint inhibition.
