Mangafodipir (Teslascan (discontinued), PledOx)
Other Medications
Description
Mangafodipir is a manganese-based contrast agent that serves as a neuroprotective compound in colorectal cancer treatment. It is specifically used as an adjunctive therapy to prevent oxaliplatin-induced peripheral neuropathy (OXAIPN) in patients receiving FOLFOX chemotherapy regimens. The drug functions as a selective antioxidant that protects normal tissues from chemotherapy-induced oxidative damage while preserving the anti-tumor efficacy of oxaliplatin-based treatments.
Mechanism of Action
Mangafodipir acts as a superoxide dismutase mimetic, catalyzing the dismutation of superoxide radicals to hydrogen peroxide and oxygen. The compound selectively accumulates in normal tissues rather than tumor cells, providing protective antioxidant activity against oxaliplatin-induced neurotoxicity without interfering with the platinum agent's cytotoxic effects on cancer cells.
