Description

Leukovorin is used in combination with fluoropyrimidine agents such as 5-fluorouracil (5-FU) or capecitabine to enhance their efficacy by providing an active form of folate that the tumor cells can use; it is commonly used in first-line and second-line treatment regimens for colorectal cancer, particularly when combined with irinotecan or oxaliplatin.

Mechanism of Action

Leukovorin acts as a folinic acid prodrug that enhances the efficacy of methotrexate by providing an active form of folic acid, which is essential for DNA synthesis and repair; it also antagonizes the toxic effects of high-dose methotrexate.
