Description
INC280 is a selective MET kinase inhibitor designed to block aberrant MET signaling in cancer cells. In colorectal cancer treatment, it is being evaluated in combination with cetuximab specifically for patients with c-MET positive metastatic colorectal cancer who have progressed on prior EGFR inhibitor therapy. The compound targets tumors that have developed resistance to standard EGFR-targeted treatments through MET pathway activation.
Mechanism of Action
INC280 functions as a selective ATP-competitive inhibitor of the MET receptor tyrosine kinase, blocking the binding of hepatocyte growth factor (HGF) and preventing downstream signaling cascades that promote tumor growth, invasion, and metastasis. By inhibiting MET signaling, the drug aims to overcome resistance mechanisms that allow colorectal cancers to continue growing despite EGFR inhibition.
