Description

GVAX is an allogeneic cellular cancer vaccine composed of irradiated colon cancer cells that have been genetically modified to secrete granulocyte-macrophage colony-stimulating factor (GM-CSF). This immunotherapy approach is designed to stimulate the patient's immune system to recognize and attack colorectal cancer cells by presenting tumor-associated antigens in an immunogenic context. GVAX is being tested in combination with immune checkpoint inhibitors like pembrolizumab and DNA methyltransferase inhibitors such as SGI-110, particularly in mismatch repair-proficient colorectal cancers that typically respond poorly to standard immunotherapy.

Mechanism of Action

The genetically modified colon cancer cells in GVAX secrete GM-CSF, which recruits and activates dendritic cells and other antigen-presenting cells at the vaccination site. These immune cells then process tumor antigens from the irradiated cancer cells and present them to T-cells, promoting a targeted immune response against similar antigens found on the patient's own colorectal cancer cells.
