Description
GTI-2040 is an experimental drug that has been studied in clinical trials for certain types of cancer, including colorectal cancer. It was investigated by the National Cancer Institute (NCI) in a Phase 1 trial, primarily to determine the highest safe dose when given in combination with standard chemotherapy drugs capecitabine and oxaliplatin. This trial focused on patients with advanced colorectal cancer or other solid tumors. The goal was to see how patients tolerated the combination and to understand how the drugs interact within the body. GTI-2040 is not currently an approved treatment for colorectal cancer.
Mechanism of Action
GTI-2040 is designed to target ribonucleotide reductase (RR), an enzyme crucial for DNA synthesis and repair in rapidly dividing cells, like cancer cells. By inhibiting RR, GTI-2040 aims to reduce the availability of building blocks needed for DNA replication, potentially slowing or stopping cancer cell growth. The trial specifically looked at changes in dCTP levels (a component of DNA) and RR-M2 mRNA levels as markers of RR inhibition.
