Description

Gad20-209-fsp is an experimental genetic vaccine being studied in clinical trials for certain types of advanced colorectal cancer. It is designed to work with the immune system to target cancer cells. Specifically, it is being investigated in patients whose tumors are MSI-H (microsatellite instability-high) or dMMR (deficient mismatch repair). These cancers have a higher number of mutations, which can make them more responsive to immunotherapy. The vaccine is given in combination with pembrolizumab (Keytruda®), an immunotherapy drug that blocks a protein called PD-1, helping the immune system attack cancer cells. The vaccine is administered as a prime shot (Gad20-209-fsp) followed by booster shots (Mva-209-fsp). It is currently being studied in Phase 1 and Phase 2 trials to determine its safety and effectiveness in this patient population.

Mechanism of Action

Gad20-209-fsp is a genetic vaccine. It uses genetic material (DNA) encoding specific peptides (Frame Shift Peptides or FSPs) found on the surface of the patient's own cancer cells. These peptides are neoantigens, meaning they are unique to the cancer and not found on normal cells. The vaccine is designed to stimulate the patient's immune system, specifically T cells, to recognize and attack these cancer-specific neoantigens. It is used in combination with pembrolizumab, which enhances the immune response against the cancer.
