Description

Doxycycline is a tetracycline antibiotic that serves as a supportive care medication in colorectal cancer treatment, specifically used to prevent and manage severe skin toxicity associated with anti-EGFR therapies like cetuximab. This broad-spectrum antibiotic is employed prophylactically to reduce the incidence and severity of acneiform rash, a common dose-limiting side effect of EGFR inhibitors. Doxycycline's anti-inflammatory properties, in addition to its antimicrobial effects, help mitigate the skin reactions that can compromise patient quality of life and treatment compliance.

Mechanism of Action

Doxycycline inhibits bacterial protein synthesis by binding to the 30S ribosomal subunit, preventing the attachment of aminoacyl-tRNA to the ribosome. Beyond its antimicrobial action, doxycycline possesses anti-inflammatory properties through inhibition of matrix metalloproteinases and reduction of inflammatory cytokine production, which helps control the inflammatory component of EGFR inhibitor-induced skin toxicity.
