Description
Docetaxel is a semi-synthetic taxane chemotherapy agent that belongs to the antimicrotubule class of cytotoxic drugs. In colorectal cancer, docetaxel is primarily used in salvage therapy for patients with metastatic disease who have progressed on standard fluoropyrimidine-based regimens. It is often combined with other agents such as gemcitabine in relapsed or refractory metastatic colorectal adenocarcinoma. Docetaxel represents a second or third-line treatment option when patients have exhausted more established chemotherapy combinations.
Mechanism of Action
Docetaxel binds to the ?-tubulin subunit of microtubules and promotes microtubule assembly while preventing depolymerization, leading to the formation of stable, non-functional microtubule bundles. This disruption of normal microtubule dynamics arrests cells in the G2/M phase of the cell cycle and ultimately triggers apoptosis. The drug's cytotoxic effect is particularly pronounced in rapidly dividing cancer cells that are more dependent on proper microtubule function for successful mitosis.
