Description
CS-1008 is a humanized monoclonal antibody that targets the death receptor 5 (DR5) pathway for apoptosis induction in cancer cells. Based on clinical trial data, it has been evaluated specifically in metastatic colorectal cancer patients who have failed first-line oxaliplatin-based treatment. The drug was tested both as monotherapy and in combination with irinotecan-containing regimens like FOLFIRI in patients with advanced disease.
Mechanism of Action
CS-1008 binds to death receptor 5 (DR5), also known as TRAIL-R2, which is expressed on the surface of many cancer cells. Upon binding, it activates the extrinsic apoptotic pathway by triggering caspase cascades that lead to programmed cell death in malignant cells while sparing normal tissues that are less sensitive to this death receptor signaling.
