Description

Cediranib (AZD2171) is an oral small molecule tyrosine kinase inhibitor that targets multiple vascular endothelial growth factor receptors (VEGFRs). In colorectal cancer treatment, cediranib has been evaluated as an anti-angiogenic agent in combination with standard chemotherapy regimens including FOLFOX for both first-line and second-line treatment of metastatic disease. The drug works by blocking tumor blood vessel formation and growth, potentially enhancing the effectiveness of cytotoxic chemotherapy.

Mechanism of Action

Cediranib selectively inhibits VEGFR-1, VEGFR-2, and VEGFR-3 tyrosine kinases, which are critical signaling pathways for angiogenesis and lymphangiogenesis. By blocking these receptors, the drug prevents the formation of new blood vessels that tumors require for growth and metastasis, effectively starving the tumor of nutrients and oxygen while also normalizing existing tumor vasculature.
