Description

Irinotecan is used in the treatment of metastatic colorectal cancer (mCRC) as a second-line therapy after fluoropyrimidine-based regimens have failed; it has shown efficacy when combined with other agents like leucovorin and 5-fluorouracil, improving overall survival rates.

Mechanism of Action

Irinotecan is a prodrug that, after being metabolized to its active form SN-38, inhibits topoisomerase I, an enzyme involved in DNA replication and transcription; this leads to DNA damage and cell death.
