Description

Calmangafodipir is a manganese-based chelate compound that functions as a neuroprotective agent specifically used to prevent oxaliplatin-induced peripheral neuropathy in colorectal cancer patients. This supportive care medication is administered alongside oxaliplatin-based chemotherapy regimens to reduce the neurotoxic effects that commonly limit treatment duration and quality of life. Calmangafodipir works by protecting peripheral nerve cells from oxidative damage caused by platinum-based chemotherapy while preserving the anticancer efficacy of oxaliplatin.

Mechanism of Action

Calmangafodipir releases manganese ions in tissues, which act as cofactors for superoxide dismutase enzymes that neutralize reactive oxygen species and reduce oxidative stress in peripheral nerve cells. The compound selectively accumulates in normal tissues rather than tumor cells, providing neuroprotection without interfering with oxaliplatin's cytotoxic effects against cancer cells.
