Description

AMG-479 is an anti-IGF-1R (insulin-like growth factor-1 receptor) monoclonal antibody designed to block IGF-1R signaling pathways that promote cancer cell growth and survival. In colorectal cancer treatment, it has been evaluated in combination with standard chemotherapy regimens like FOLFIRI, particularly for patients with KRAS-mutant metastatic disease. The drug targets a pathway involved in tumor progression and metastasis in various solid tumors including colorectal carcinoma.

Mechanism of Action

AMG-479 binds specifically to the insulin-like growth factor-1 receptor (IGF-1R) on cancer cells, preventing IGF-1 and IGF-2 from activating this receptor. By blocking IGF-1R signaling, the antibody inhibits downstream pathways including PI3K/AKT and MAPK that promote cancer cell proliferation, survival, and resistance to therapy.
