Description

ABX-EGF is the developmental code name for panitumumab, a fully human monoclonal antibody that targets the epidermal growth factor receptor (EGFR). This targeted therapy is specifically used to treat metastatic colorectal cancer in patients whose tumors express EGFR and do not have KRAS mutations. Panitumumab is administered as a monotherapy or in combination with chemotherapy regimens for patients with advanced colorectal carcinoma.

Mechanism of Action

Panitumumab binds specifically to the extracellular domain of the human EGFR, blocking the binding of natural ligands such as EGF and transforming growth factor-alpha. This binding inhibits EGFR autophosphorylation and downstream signaling pathways including RAS-MAPK and PI3K-AKT, ultimately leading to inhibition of cell growth, induction of apoptosis, and decreased angiogenesis in EGFR-expressing tumor cells.
