Description
Trastuzumab is a monoclonal antibody that targets HER2-positive colorectal cancer, a rare subtype representing approximately 2-5% of all CRC cases. In metastatic HER2-positive colorectal cancer, trastuzumab is used in combination with other agents such as tucatinib and chemotherapy regimens like mFOLFOX6 as first-line treatment. The drug has shown efficacy in HER2-amplified colorectal tumors, particularly when combined with other HER2-targeted therapies in dual HER2 blockade strategies.
Mechanism of Action
Trastuzumab binds specifically to the extracellular domain of the HER2 receptor (human epidermal growth factor receptor 2), blocking HER2-mediated cell signaling pathways that promote cancer cell growth and survival. By binding to HER2, it prevents receptor dimerization and downstream activation of PI3K/AKT and MAPK pathways, leading to reduced tumor cell proliferation and enhanced apoptosis. The antibody also recruits immune effector cells through antibody-dependent cellular cytotoxicity (ADCC).
