Description

Spartalizumab (PDR001) is a humanized monoclonal antibody that functions as an immune checkpoint inhibitor targeting the PD-1 receptor. In colorectal cancer, it has been evaluated specifically in combination with regorafenib for previously treated metastatic disease and as part of combination regimens for BRAF V600-mutated tumors. The drug is designed to enhance the immune system's ability to recognize and attack cancer cells by blocking inhibitory signals that tumors use to evade immune detection.

Mechanism of Action

Spartalizumab binds to the programmed cell death protein-1 (PD-1) receptor on T-cells, preventing the interaction between PD-1 and its ligands PD-L1 and PD-L2. This blockade removes the inhibitory brake on T-cell activation, allowing immune cells to maintain their anti-tumor activity. The enhanced T-cell response can lead to increased tumor cell death and improved immune surveillance against cancer.
