Description
Sintilimab is a humanized monoclonal antibody that functions as a PD-1 immune checkpoint inhibitor used in colorectal cancer treatment. It is primarily utilized in patients with microsatellite instability-high (MSI-H) or mismatch repair deficient (dMMR) colorectal tumors, where it demonstrates enhanced efficacy due to the high mutational burden characteristic of these cancers. The drug has been evaluated in both stage III resectable disease as neoadjuvant therapy and in combination regimens with other agents like bevacizumab for microsatellite stable (MSS) tumors.
Mechanism of Action
Sintilimab binds specifically to the programmed death-1 (PD-1) receptor on T lymphocytes, blocking the interaction between PD-1 and its ligands PD-L1 and PD-L2. This blockade removes the inhibitory signal that normally suppresses T-cell activation, thereby restoring and enhancing the immune system's ability to recognize and destroy cancer cells.
