Description

Rivaroxaban is a direct oral anticoagulant (DOAC) that belongs to the factor Xa inhibitor class of medications. In colorectal cancer treatment, rivaroxaban serves as supportive care for the prevention and treatment of cancer-associated thromboembolism, a serious complication that affects up to 20% of cancer patients. It is also used for extended antithrombotic prophylaxis following laparoscopic colorectal cancer surgery to reduce the risk of venous thromboembolism during the postoperative recovery period.

Mechanism of Action

Rivaroxaban directly and selectively inhibits factor Xa, a key enzyme in the coagulation cascade that converts prothrombin to thrombin. By blocking factor Xa, rivaroxaban prevents thrombin generation and fibrin clot formation, thereby reducing the risk of both venous and arterial thrombotic events. This mechanism is particularly important in cancer patients who have an increased baseline risk of thrombosis due to tumor-related procoagulant factors and treatment-related hypercoagulability.
