Description

Pertuzumab is a humanized monoclonal antibody that targets HER2-positive colorectal cancers, used in combination with trastuzumab for patients whose tumors overexpress or amplify the HER2 protein. In colorectal cancer, pertuzumab is primarily utilized for HER2-positive metastatic disease, which occurs in approximately 2-3% of colorectal cancer patients. This targeted therapy represents a precision medicine approach for a specific molecular subset of colorectal cancer patients who have limited treatment options.

Mechanism of Action

Pertuzumab binds to the extracellular dimerization domain of the HER2 receptor, preventing heterodimerization with other HER family receptors including HER1, HER3, and HER4. By blocking HER2 receptor dimerization, pertuzumab inhibits downstream signaling pathways that promote tumor cell proliferation and survival. When combined with trastuzumab, pertuzumab provides complementary HER2 blockade, as trastuzumab binds to a different epitope on the HER2 receptor.
