Description
Pazopanib is an oral multi-targeted tyrosine kinase inhibitor that blocks several growth factor receptors involved in tumor growth and angiogenesis. While FDA-approved for renal cell carcinoma and soft tissue sarcoma, pazopanib has been explored in colorectal cancer through combination regimens, particularly as a second-line treatment option combined with irinotecan and cetuximab. The drug's anti-angiogenic properties make it a rational candidate for CRC treatment, where blood vessel formation is critical for tumor progression and metastasis.
Mechanism of Action
Pazopanib inhibits multiple receptor tyrosine kinases including VEGFR-1, VEGFR-2, VEGFR-3, PDGFR- , PDGFR- , and c-Kit, which are essential for angiogenesis and tumor cell proliferation. By blocking these pathways, pazopanib disrupts the formation of new blood vessels that feed tumors and interferes with growth signals that promote cancer cell survival and division.
