Description
Panobinostat is a pan-histone deacetylase (HDAC) inhibitor that has been evaluated in colorectal cancer treatment, particularly in combination with fluorouracil-based regimens for metastatic disease. This epigenetic modifier works by altering gene expression patterns in cancer cells and has been studied in patients with stage IV colorectal cancer who have not responded to previous fluoropyrimidine therapy. Panobinostat represents part of combination approaches that include immunotherapy agents like PDR001 and other targeted therapies for advanced colorectal cancer.
Mechanism of Action
Panobinostat inhibits multiple classes of histone deacetylases (HDACs), enzymes that remove acetyl groups from histone proteins and regulate gene transcription. By blocking HDAC activity, panobinostat increases histone acetylation, leading to chromatin relaxation and reactivation of tumor suppressor genes that are silenced in cancer cells. This epigenetic modification can restore normal cell cycle control and promote cancer cell death.
