Description
Napabucasin (BBI-608) is a small molecule STAT3 pathway inhibitor that has been evaluated for treatment of metastatic colorectal cancer. This oral agent targets cancer stem cell pathways and has been studied in combination with standard chemotherapy regimens like FOLFIRI for patients with previously treated metastatic disease. The compound also shows potential for combination with immunotherapy agents like pembrolizumab.
Mechanism of Action
Napabucasin inhibits the STAT3 (Signal Transducer and Activator of Transcription 3) signaling pathway, which is frequently activated in cancer stem cells and promotes tumor growth, metastasis, and treatment resistance. By blocking STAT3 activity, the drug aims to target the cancer stem cell population that can drive tumor recurrence and progression.
