Description
Mangafodipir (mangafodipir trisodium) is a manganese-based chelate compound used as a supportive care agent to prevent oxaliplatin-induced peripheral neuropathy in colorectal cancer patients receiving FOLFOX chemotherapy regimens. The drug serves as a neuroprotective agent specifically designed to reduce the neurotoxic side effects of oxaliplatin while preserving the chemotherapy's anti-cancer efficacy. Mangafodipir has been evaluated in both adjuvant and metastatic colorectal cancer settings as an adjunct to standard FOLFOX6 treatment protocols.
Mechanism of Action
Mangafodipir acts as a selective scavenger of reactive oxygen species and functions as a superoxide dismutase mimetic, particularly protecting neuronal tissues from oxidative damage. The compound preferentially accumulates in normal tissues rather than tumor cells, allowing it to provide neuroprotection without interfering with oxaliplatin's cytotoxic effects against cancer cells.
