Description

Ivosidenib is approved for the treatment of patients with IDH1-mutant relapsed or refractory acute myeloid leukemia (AML) and has shown efficacy in certain solid tumors, including some cases of colorectal cancer. However, its use in colorectal cancer remains investigational and limited to clinical trials evaluating its role in specific subtypes.

Mechanism of Action

Ivosidenib is a selective inhibitor of mutant isocitrate dehydrogenase-1 (IDH1) and acts by inhibiting the production of 2-hydroxyglutarate, which leads to decreased DNA hypermethylation and cell differentiation.

Fatigue; nausea; diarrhea; vomiting; constipation; abdominal pain; decreased appetite; rash; myalgia; arthralgia
