Description

ISIS refers to antisense oligonucleotide compounds developed by Ionis Pharmaceuticals (formerly ISIS Pharmaceuticals) that have been evaluated in colorectal cancer treatment. ISIS 2503 targets H-ras mRNA to reduce oncogenic H-ras protein production, while ISIS 183750 targets protein tyrosine phosphatase 1B (PTP1B) and has been combined with irinotecan in advanced colorectal cancer. These antisense oligonucleotides are designed to bind to specific mRNA sequences and prevent translation of proteins that promote cancer cell growth and survival.

Mechanism of Action

ISIS 2503 works by binding to H-ras mRNA through complementary base pairing, leading to mRNA degradation and reduced production of the H-ras oncoprotein that drives cell proliferation. ISIS 183750 targets PTP1B mRNA, reducing PTP1B protein levels which normally inhibit insulin and growth factor signaling pathways involved in cancer progression.
