Description

Indomethacin is a nonsteroidal anti-inflammatory drug (NSAID) that belongs to the indole acetic acid derivative class. In colorectal cancer, it is used as supportive care medication and has been explored in combination with standard chemotherapy regimens including fluorouracil and platinum-based treatments. The drug serves both as an anti-inflammatory agent for symptom management and has shown potential anticancer properties in early-phase clinical studies. Its use in CRC is primarily off-label, as it is not specifically approved for cancer treatment.

Mechanism of Action

Indomethacin primarily inhibits cyclooxygenase (COX) enzymes, particularly COX-1 and COX-2, which reduces prostaglandin synthesis and inflammatory responses. In the cancer context, COX-2 inhibition may help reduce tumor-promoting inflammation and potentially enhance the efficacy of concurrent chemotherapy agents. The drug may also have direct antiproliferative effects on cancer cells through prostaglandin-independent pathways.
