Description
HRS-7058 is a selective KRAS G12C inhibitor designed to target colorectal cancers harboring specific KRAS mutations. This small molecule therapy is being evaluated in neoadjuvant treatment protocols for locally advanced colorectal cancer as part of umbrella trial designs. The drug specifically binds to the KRAS G12C mutant protein, which is found in approximately 3-5% of colorectal cancer patients and represents a historically difficult-to-target oncogenic driver.
Mechanism of Action
HRS-7058 works by covalently binding to the cysteine residue in the KRAS G12C mutant protein, locking it in an inactive GDP-bound state. This prevents the mutant KRAS protein from transmitting growth signals through downstream pathways including RAF-MEK-ERK and PI3K-AKT, thereby inhibiting tumor cell proliferation and survival.
