Description

Entrectinib is used in the treatment of NTRK fusion-positive solid tumors and ROS1-positive non-small cell lung cancer (NSCLC); it has shown efficacy in patients with advanced or metastatic cancers; however, its use in colorectal cancer (CRC) remains investigational as of now.

Mechanism of Action

Entrectinib is a selective, potent inhibitor of tropomyosin receptor kinase (TRK) A/B/C and ROS1 kinases; it binds to the ATP-binding site of these kinases, thereby inhibiting their activity and downstream signaling pathways.

Side Effects

Hemorrhage; fatigue; constipation; dysgeusia; dizziness; anemia; nausea; vomiting; increased AST levels; increased ALT levels
