Dasatinib (Sprycel)
Targeted Therapy · Approved since 2006
Description
Dasatinib is a multi-targeted tyrosine kinase inhibitor that has been evaluated for treatment of metastatic colorectal cancer in patients with specific molecular profiles. This oral targeted therapy agent is primarily FDA-approved for certain blood cancers but has been studied in solid tumors including CRC through precision medicine approaches. In colorectal cancer, dasatinib has been explored as a treatment option for patients with refractory disease, particularly in the context of personalized treatment strategies based on tumor genetic testing.
Mechanism of Action
Dasatinib works by blocking multiple tyrosine kinases including BCR-ABL, SRC family kinases, c-KIT, and platelet-derived growth factor receptors. By inhibiting these signaling pathways, particularly SRC kinases that play roles in tumor cell proliferation, invasion, and metastasis, dasatinib can potentially slow cancer growth and spread in colorectal tumors with appropriate molecular characteristics.
