Description
Celecoxib is a selective cyclooxygenase-2 (COX-2) inhibitor that belongs to the class of nonsteroidal anti-inflammatory drugs (NSAIDs). In colorectal cancer, celecoxib is used as adjuvant therapy alongside chemotherapy in metastatic disease and for chemoprevention in patients with familial adenomatous polyposis (FAP). The drug has demonstrated anti-tumor properties and is employed to reduce adenoma formation and potentially enhance the effectiveness of standard cancer treatments.
Mechanism of Action
Celecoxib selectively inhibits cyclooxygenase-2 (COX-2), an enzyme that catalyzes the conversion of arachidonic acid to prostaglandins. By blocking COX-2, celecoxib reduces the production of prostaglandin E2 (PGE2), which promotes tumor growth, angiogenesis, and metastasis in colorectal cancer. This selective inhibition also decreases inflammation in the tumor microenvironment while potentially enhancing immune responses against cancer cells.
