Description

Camrelizumab is a humanized monoclonal antibody that functions as a PD-1 (programmed death-1) immune checkpoint inhibitor used in colorectal cancer treatment. In CRC, it is primarily employed in combination regimens for patients with microsatellite instability-high (MSI-H) tumors and as part of multimodal treatment approaches for locally advanced rectal cancer. The drug has been evaluated in combination with chemotherapy agents like capecitabine and targeted therapies such as fruquintinib for patients with unresectable metastatic disease. Clinical applications include both microsatellite stable (MSS) and MSI-H colorectal cancers, often integrated with radiation therapy and chemotherapy protocols.

Mechanism of Action

Camrelizumab binds specifically to the PD-1 receptor on T-cells, blocking the interaction between PD-1 and its ligands PD-L1 and PD-L2. This blockade prevents the inhibitory signals that normally suppress T-cell activation, thereby restoring and enhancing the immune system's ability to recognize and attack cancer cells. By removing this immune checkpoint brake, camrelizumab enables sustained anti-tumor immune responses.
